Police said the bus, apparently full of commuters, was hit by the train at about 9am outside the town of Marhanets, in the Dnipropetrovsk region.
The bus had driven through a red light at a level-crossing, ignoring a siren that indicated oncoming trains, according to reports.
"The driver of the bus broke traffic laws by driving through a red light before colliding with the train," said Lyudmila Bashmakova, a spokeswoman for the regional traffic police.

"It's utter carnage there."
Two children were among the dead.
"The motor of the bus stopped at the level crossing as the driver was changing gears," Ukraine's interior ministry said, quoting witnesses.
Officials said the crossing, which operated with a sound and light system, was automatic and in good working order.
"Survivors remember yelling (to the driver) that the sound signal means he cannot proceed," Ukrainian Transportation Minister Kostyantyn Efimenko said.
